Tato práce si kladla za cíl prostudovat možnosti optimalizace nanovlákenných struktur s inkorporovaným SNAPsem, jenž zastává funkci donoru oxidu dusnatého. Byl zkoumán jeho vliv na morfologickou stavbu nanovlákenných nosičů vytvořených z alifatických polyesterů PCL a PLC (PURASORB) pomocí elektrostatického zvlákňování a rovněž je zkoumán vliv SNAPsu na buněčnou adhezi a viabilitu. Dále byly prostudovány možnosti detekce uvolněného oxidu dusnatého z nanovlákenné struktury pomocí nepřímé metody Griess Assay a vliv sterilizace pomocí 70 hm % etanolu na buněčnou viabilitu. Na základě sérií testování byl vyhodnocen nejvhodnější materiál spolu s koncentrací SNAPsu, který je nejvíce vyhovujícím pro vytvoření maloprůměrové cévní náhrady.

Purpose of this work was study of optimisation possibilities concerning nanofibrous structures with incorporated SNAP, which acts as a donor of nitric oxide. His influence on the morphological structure of nanofibrous carriers made of aliphatic polyesters PCL and PLC (PURASORB) using electrostatic spinning, as well as the effect of SNAP on cell adhesion and viability. Furthermore we explore the possibilities of detection of released nitric oxide from the nanofibrous structure by the indirect Griess Assay method and the effect of sterilization with 70 wt % etanol on cell viability. Based on series of testing, the most suitable material was evaluated, together with the concentration of SNAP, which is most suited for the creation of a small diameter vascular substitute.
